Depends on whether the '00 has "C" nav or "D" nav. If "C" nav, no. If "D" nav, yes. The W220 S-Class Encyclopedia sticky has links that tell you how to determine which system is installed in the '00.

I believe it won't work because the 2003 uses DVD and the 2001 uses CD based navigation.
You are correct in your last post. The '03 W220 does not use nav DVDs; the COMAND MOPF is wide screen, but it is still CD based, and uses the same CDs as the '00 "D" nav system as well as the '01 and '02 models.

One nice thing about the "D" nav sets (since the '03 CD update, as I recall) is that they have the entire U.S. interstate highway system on every disc. The maps cover major roads for about a mile either side of the interstate. This means, for example, that you can start from any interstate segment in San Francisco, with the nav disc for Miami, and drive the interstates to Miami without changing discs; and you'd have coverage sufficient to handle motels and restaurants along the way. Not as good as the DVDs, but still a reason to keep the entire CD set.
