What's the difference between the S550 and S550V is one a long wheel base or something.

Is the short wheelbase model available in Canada? V refers to the long wheelbase in the chassis models number, like V221 is the LWB car and a W221 is the SWB model ("unofficially" W211 is often used for both though).

I've never seen this "S550V" but in the US they name, was it E350S and E350W for wagon (S) and sedan (W) which come from the same body naming approach, W211 for sedan and S211 for wagon.

As far as I know, the S550 name is for North American bound S classes only. Mercedes is only sending long wheelbase cars here. Technically, all S550 are S550V, because V designates long wheelbase.
