Same problem I had with the S600 last year; the key wouldn't turn at all. There are a few things to check.

1.) Did the fuse that powers the EIS (the ignition key receptacle) go pop? That's what happened on mine.
2.) Did the EIS itself go bad? There are US $10.00 things called EIS testers that will see if you're getting current flow through it.
3.) Do you have more than one Mercedes-Benz, with similar-looking keys, and put the wrong key into it? Don't laugh too hard; I've done this more times than I care to admit Granted, that's kind of a good "problem" to have.

If it's the fuse popping, then time to figure out why. In my case, the reed valve up front had gotten plugged, water got into the passenger-side interior, and things started shorting out (I fixed it).

First check is always, "Is the battery really good?".

I discovered that if the battery has at least 11.5V, the key will turn. Didn't try going lower than that and not going to...not purposefully, anyway. :-)

First, fully recharge the battery and have it tested. Then, determine why it went dead, and fix that.

The Encyclopedia of Stickies has the fuse charts for all models of W220, by VIN.

Including Pre Fuse Locations

Of course the first thing I'd do is plug it into SDS and see what is or isn't going on

SDS - as best as I can tell - requires the key to be in the ACC position to work.
