Only way know for sure is to make an offer and start the negotiation process. You need to find the car you want first though. Negotiation can only be done for real.
Depending on dealer and discounts (fleet?) for which you are eligible, I have been quoted way below invoice. Also depends on which car you want. Lots of sport models out there, but very few luxury sedans, at least in the Southeast.
Just ordered a E350 4 Matic for Feb delivery. MSRP was just about 60K. The 54,145 must be a lightly equipped car. Mine was discounted but I do not disclose any prices here or elsewhere in public.

Because there is so much misleading price info on these sites. Prices vary from region to region and from dealer to dealer depending on their situation. In many cases the price given here does not have the full story..ie demo car, fleet discount, employee discount, etc. etc. Some others just blatently mis state the price to the point where it makes no sense at all. Anyone capable of purchasing an MB should be able to negotiate the best deal themselves. I am not going to jump into that mess!
