I picked up my new ML320 cdi sport one week ago (also in Yorkshire) and I've got exactly the same problem. For me it feels like the top of the lumbar support is pressing into my back even when fully off causing me loads of discomfort and back ache after only a few minuts of driving.
My dealer says he will happily take the lumbar support out and replace it with foam and will even more happily charge me 500 quid for the privilege.
Have you had any more luck in getting more comfortable? Does anyone else have any suggestions other than "you'll probably just get used to it"?
It does take a bit of seat adjustment to get "comfortable" or in what feels like the best driving position in the ML.
What I really don't like is the bulky oversized steering wheel with the fake plastic spokes at the bottom. Compared to the proper steering wheel in the 08 C-class, it's junk.

At 6'1" (and shrinking) I usually drive a Ford F150 pickup truck and like seating position so when we got the ML for my wife I adjusted the seat similarly. But since the seat does not lower (like in our last Infiniti) I banged my head every time I got into it. So now the memory is set but I have to remember to lower the seat before getting in and then select my setting. On longer drives I really like the comfort factor but my wife has issues with the seating and that has been the case with the last five cars we've had. I've bought her three different types of cushions and nothing fits her butt so I've given up and turned on my selective hearing filter.
Actually in re-reading this thread I think the comfort problem is only with ML's sold in Yorkshire. The steering wheel is probably on the wrong side, or M-B forgot to swap the drivers seat from the left to right
