Not specifically for the ML to my knowledge, so the S-class's is not necessarily specifically designed for the weight of the ML's wheels. Those ML55 ones can be pretty darn heavy!

Is there any clearance issues with them on a ml320, and have people who have done this found that they stick out to far without flares.

No, they fit fine. They stick beyond the fenders by a little (since the tire is 30 millimetres wider than the ML320's stock), but certainly not too far.
