As others have mentioned, the top things you need to do are:
1) Drive slower. This makes the biggest difference, by far. Accelerate slowly at lights and keep it to the speed limit. Don't use cruise control. Your gas mileage can vary up to 10mpg (from personal experience) depending on how you drive.
2) Fill your tires to just under sidewall pressure. It may affect safety and ride but it will make a 2-4mpg difference in gas mileage.
3) Stop using ethanol gas! Get the good stuff, 93 octane pure gasoline. Another 3-5mpg difference.
4) If you haven't already - keep up on regular maintenance. Change the air filter and the oil. Depending on how dirty they are this may add another 1-2mpg.
Things not to worry about:
Air conditioning. It makes no difference.
Shedding weight. Unless you are carrying around hundreds of pounds of crap it isn't going to make a difference. The weight of the spare tire won't affect your gas mileage at all.
Fuel additives - no, just no.
